Hi,
Can anyone tell me what the calibre of the 20mm and 37mm cannons on the S-100 boats were so that I can replace the barrels.

Hello Toby, I believe both the 20mm, and the 3.7cm guns used on the S-Boote were the same as the ones used by the German army as Flak guns. Any of the barrels made for the Wehrmacht versions of those guns should work. The four barreled Flakvierling is the same gun(s) and mount used by both the Wehrmacht and the Kriegsmarine.
